ABP Biosciences 公司最新推出一系列生物素标记的dCTP, dATP, dUTP和UTP探针以及荧光素标记的dUTP和UTP探针。这些探针可广泛应用于Hi-C 测序基因捕获, FISH, TUNEL, DNA microarray 等应用。其中,荧光素标记的dUTP和UTP探针具有荧光亮度强,光稳定性好等优点,另外探针在染料和dUTP及UTP之间含X-linker以确保在酶的作用下更高效的插入DNA或RNA。

图1. FISH探针的应用。Human Chromosome 7 Enumeration (CEP7) 和 Human c-Met gene (MET) 探针采用 nick translation 技术分别标记Andy Fluor 488-X-dUTP (绿色) 和 Andy Fluor 568-X-dUTP (红色)。标记后的CEP7/ MET探针与HT1080细胞在37ºC孵化过夜,经洗涤后用抗氧化封闭液封片,然后在荧光显微镜下成像。

图2. 细胞凋亡TUNEL检测的应用。小鼠舌组织的凋亡细胞利用TUNEL技术在TdT酶的催化下标记Biotin-11-dUTP,然后用 Andy Fluor™ 488标记的streptavidin显色(左图) 或者采用 horse radish peroxidase (HRP) 标记的streptavidin/DAB 显色(右图)。
