DNA含量的测定可以用来研究细胞在各阶段周期的分布以及DNA倍体的分析。在给定群体中，细胞主要分布在细胞周期的三个阶段：G0 / G1期，S期和G2 / M期。在G2 / M期，细胞含双份基因组DNA，是G0 / G1期细胞的2倍，而正在进行DNA复制的S期细胞的DNA含量介于二者之间。当选用DNA选择性染料来测量DNA含量时，其荧光强度与DNA含量成正比。细胞内的DNA被染色后，可以用流式细胞仪对细胞进行DNA含量测定，然后根据DNA含量的分布情况，可以进行细胞周期分析。
