NGS Quick Ligation Module

The NGS Quick Ligation Module is optimized to ligate DNA adaptors compatible with Illumina sequencing to end-repaired, dA-tailed DNA fragments. The module is optimized for use with the NGS End Repair Module (Cat #E112-1, E112-2) and the NGS dA-Tailing Module (Cat #E113-1, E113-2) and is part of the NGS DNA library prep workflow, which is suitable for 1–5 μg of input DNA.

Applications

• Ligation of DNA adaptors to blunt or dA-Tailed DNA.
• Ligation of blunt or dA-Tailed DNA to cloning vectors.
• DNA sample preparation.

Protocol 

1. Assemble the following reaction in a microcentrifuge tube on ice:

     End Repaired, Blunt or dA-Tailed DNA           1-5 µg
     Quick Ligation Reaction Buffer (5×)                10 µL
     DNA Adapters                                                  variable
     Quick T4 DNA Ligase                                      5 µL
     Nuclease-free water                                        to 50 µL
    Total volume                                                    50 µL

2. Mix gently and spin down for a few seconds.

3. Incubate at 20°C for 30 min.

4. Purify DNA Sample using AMPure XP beads.
